چکیده

THE separation and purification of human islets from the pancreas of pediatric-age donors has been considered difficult if not impossible because of easy fragmentation of the islets and/or their incomplete separation from the enclosing acinar tissue (embedding). I.:! The purpose of this study was to develop a method to separate and purify large numbers of viable human islets from pediatric donors. After preliminary studies on swine islet isolation3 which resemble pediatric human isolation in many ways, a modification of the automated method for adult human islet isolation was developed.

Pancreas procurement from multiorgan donors for islet trasplantation.

The outcome of human islet isolation procedures can be significantly effected by the technique used for pancreas procurement. In fact, the final step of islet purification using discontinuous density gradients requires a significant difference between the density of the islets and the density of the non-endocrine component of the gland. Therefore, any procedure during multi-organ procurement th...

Successful human islet isolation utilizing recombinant collagenase.

The enzymatic dissociation of acinar tissue by collagenase is a substantial step in the isolation of pancreatic islets. Although essential collagenase components have been purified, the variability in the activity of different batches limits long-term reproducibility of isolation success. The utilization of purified recombinant proteases would solve this problem. In the present study, pancrease...

Islet Transplantation in Diabetes: the Pittsburgh Experience

Nineteen-ninety was a significant year for clinical islet transplantation, since after many attempts reports of short terml and prolonged2.6 insulin independence following human islet allotransplantation, indicated that it is possible to replace the endocrine function of the pancreas by an islet transplant in man. The development of improved procedures for islet isolation and purification from ...

Improvement of pancreatic islet cell isolation for transplantation.

Pancreatic islet transplantation is a promising treatment for diabetes but still faces several challenges. Poor islet isolation efficiency and poor long-term insulin independence are currently two major issues, although donor shortage and the need for immunosuppressants also need to be addressed. We established the Kyoto islet isolation method (KIIM), which has enabled us to isolate and transpl...
